You'll need to 'assign' each individual sound file to each individual message event under F3 Options -> Messages. You can, if you so desire, assign the same sound file to different message events.

It shouldn't matter which simulation you open up to set up the sound files, as all or almost all message events are universal across every simulation. It also shouldn't matter where the sound file itself is stored, as long as it is accessible to your system as you are playing a simulation. Personally speaking, I've created a "Sounds" folder in that same folder you are currently in in your screenshot to keep all of my sound files together in one place.

Reading through this previous thread may be of interest and use to you with regards to setting up sound files. I wrote an extensive post in that same thread detailing and linking to which sound files I've attached to each message event and why.
