I am running the 2012 timetable, whenever I have anything going north out of Temple Meads, heading towards Filton it always stops a signal 57 and complains about wrong route being set. This applies to any of the possible combinations of route past there. Trains coming out of plat 3 on to the relief aren't affected as they don't go past 57.

Is the simulation definitely running in the 'Modern' era (i.e. can you see platforms 13 and 15 at Bristol)? That's the only thing I can think of, trains to Filton should accept the route from 57 to either 131 or 231.

If you're running the correct era, please could you upload a save? I've never encountered this problem before.

I seem to have solved it. It seems that I was incorrectly sending the route through 569 when it is possible to send a train straight through there without including it in the route.

Yep, wherever possible you should set routes between main signals only. Set via shunt signals only if there's no other option (e.g. at St Phillips Marsh) or you're shunting freights, locos or ECS.
