With certain simulations already released, you can put BLOK on the first available berth and the sim automatically diverts any service booked to use that line onto a alterative line ie Swindid.

Now I'm playing Rugby South and have a TCF between signal KR5262 and signal KR5260 now my question as I can't seem to find the answer in the manuals available is in the berth which shows approaching trains, if I put BLOK there will the sim pick this up and divert services booked to use the Up Slow from Hanslope Junction to instead use the Up Fast instead then cross back over to the Up Slow at Denbigh Hall North Junction instead?

BLOK codes have been set up on the Rugby simulations. You're not allowed to interpose into queue berths (which is what the 'APPR' berth is), but if you insert 'BLOK' at the first signal you control (KR5268 in this case), trains will divert to the Up Fast.

Does that apply to all the Rugby sims at the first controlled signal of the revlant sim so say you wanted a block of the Up and Down Fast lines between Watford Jcn and Tring you could?

I've added to the Rugby South and Centre manuals the appropriate signals for blocking. I have to wait for Noel to get back to do North.

Generally it's the first signal supervised by the signalbox being simulated (automatic or controlled signal).

Is it possible to also implement it so that using the line in the opposite direction also behaves in the same way as the BLOK code, ie so that using the Up Fast between Brinklow and Nuneaton as a down line will force everything to use the slows? I tried once and the trains just waited until I gave the line back to the up direction.

Just interpose BLOK into berth 3382. It will not get overwritten by down trains, and up trains will use the slow.
